Effects of L-3,4-dihydroxyphenylalanine (L-Dopa) Treatment in the Neuroendocrine Response to Stress

چکیده
Abstract Stressful stimuli evoke a complex response mediated by two systems: the Sympathetic-Adreno-Medullar (SAM) axis and Hypothalamus-Pituitary-Adrenal (HPA) axis. Among factors involved in stress, glucocorticoids catecholamines secreted from adrenal glands sympathetic nerves are main effectors of physiological adaptations to stressors. Besides these, prolactin (PRL) is another hormone under stress conditions. Catecholamines synthesized hydroxylated precursor L-Dopa. This agent commonly used for treatment Parkinson’s disease it would act as neurotransmitter per se. On other hand, has been suggested that HPA dysregulation potential risk factor development depression. In line with this, several studies reported L-Dopa may alter serum levels ACTH, PRL, parkinsonian patients animal models. present study, we determined whether chronic altered inducing depressive-like behaviours. Adult male Wistar rats were treated orally during 24 days LEBOCAR® - commercial formulation (75 mg/day) Carbidopa (7.5 drinking water. Animals stressed immobilization last 9 behaviours assessed sucrose intake forced swimming tests. Behavioural tests showed no signs LEBOCAR®-treated and/or rats. We next explored SAM reactivity. Circulating noradrenaline adrenaline increased (p<0.05; HPLC). Also, adrenals animals higher content (p<0.05). Then, studied activity. Chronically displayed lower ACTH secretion (ELISA) downregulation POMC expression (qPCR) anterior pituitary addition, induced reduction As expected, corticosterone enhanced an effect was inhibited Finally, PRL gene downregulated more pronounced when also Our results suggest alters neuroendocrine enhancing reactivity reducing activity expression.

Continuous and intermittent nicotine treatment reduces L-3,4-dihydroxyphenylalanine (L-DOPA)-induced dyskinesias in a rat model of Parkinson's disease.
The development of abnormal involuntary movements (AIMs) or dyskinesias is a serious complication of L-DOPA [L-3,4-dihydroxyphenylalanine] therapy for Parkinson's disease. Our previous work had shown that intermittent nicotine dosing reduced L-DOPA-induced dyskinetic-like movements in nonhuman primates. A readily available nicotine formulation is the nicotine patch, which provides a constant so...

OBJECTIVE To determine whether a positive L-dopa response in vascular parkinsonism (VP) is correlated with the presence of nigrostriatal pathology due to either vascular damage or neuronal cell loss. METHODS Seventeen patients with pathologically confirmed VP were selected from the pathological collection of the Queen Square Brain Bank for Neurological Disorders, and their L-dopa response dur...
متن کاملActivation of dopamine cell firing by repeated L-DOPA administration to dopamine-depleted rats: its potential role in mediating the therapeutic response to L-DOPA treatment.
The administration of L-dihydroxyphenylalanine (L-DOPA) to patients with Parkinson's disease is known to produce acute effects that include the reduction of rigidity as well as delayed therapeutic actions involving the resumption of complex motor behavior. In order to examine the potential role of dopamine (DA) cell activity in mediating these responses, the effects of acute and repeated L-DOPA...

متن کاملBehavioral and anatomical effects of long-term L-dihydroxyphenylalanine (L-DOPA) administration in rats with unilateral lesions of the nigrostriatal system.
This study investigated behavioral and anatomical changes induced by long periods of L-DOPA treatment in the unilateral rat model of Parkinson's disease. After daily injections of L-DOPA (50 mg/kg, ip) given for 1, 4, 8, or 16 weeks, behavioral sensitization, expressed by contralateral turning and changes in its pattern, increased within the first week of treatment and remained unchanged therea...
